Dear Stephane,

Thank you very much for your swift reply! It was incredibly helpful and I have successfully made the workflow using your suggestions. One other problem I ran into was the “Filter ROI” block. I am trying to filter ROIs detected using the “wavelet spot detector” based on MaxIntensity. I unfortunately keep running into the following error:

java.lang.UnsupportedOperationException: ROIMaxIntensityDescriptor: cannot compute descriptors for ‘spot #8526’
Caused by :
java.lang.UnsupportedOperationException: Not allowed to cannot compute intensity descriptor on a multi channel ROI (sizeC > 1).

I am slightly confused, as I am using the following  inputs for the “Filter ROI” block: ROIs : ‘detection as ROIs‘ from “Wavelet Spot Detector” Block Sequence: ‘Extracted‘ from “Extract Channel” block, so there should only be 1 channel for each  ROI correct. Is there a intermediate step I am missing?

Best,

Katie
